Proteins are complex mixtures and can be in a variety of states in the cell. To separate one particular protein from other proteins, cells are broken open using methods such as mechanical disruption (cell lysis), chemical lysis with detergents and enzymes or sonication. The resulting mixture is called the lysate and it contains the target protein along with other cellular components.
